Properties
It is a short-acting sulfonamide derivative that develops chemotherapeutic-antibacterial activity on numerous gram-positive and gram-negative pathogenic microorganisms. They are absorbable sulfonamides that act as antimetabolites (antifolates) by combining with the same enzyme that acts on PABA to transform it first into dihydrofolate and then into tetrahydrofolate through a mechanism of competition for the substrate. Their digestive absorption varies from one derivative to another, they diffuse through the blood and tissues, and are eliminated mainly in urine as intact drug or acetylated derivatives. Its half-life is 8 hours. Its main biotransformation process (acetylation) occurs mainly in the liver, and the acetylated fraction appears in the blood plasma (25-30%). With the advent of antibiotics, sulfonamides have lost therapeutic interest since they have been widely surpassed in effectiveness and tolerance.

Dose
Sulfadiazine is used orally combined with tetroxoprim (100mg/250mg) every 12 hours. For topical use it is associated with allantoin, menthol, benzocaine, chlorophyll, etc., in oropharyngeal mouthwashes for angina, pharyngitis and tonsillitis. As a single drug, 2g is administered in the first dose and then 1g every 6 hours.

Side effects
Occasionally, nausea, vomiting, epigastralgia, fever, headaches, pruritus, skin reactions, leukopenia, thrombocytopenia, biliary cholestasis, nephropathies, thrombocytopenic purpura, epidermal necrolysis, hepatitis, hemolytic anemia, crystalluria.
